چکیده
Abstract Background Muscle is severely affected by ischemia/reperfusion injury (IRI). Quiescent satellite cells differentiating into myogenic progenitor (MPC) possess a remarkable regenerative potential. We herein established model of local application MPC in murine hindlimb to study cell engraftment and differentiation required for muscle regeneration. Methods A clamping (C57b/6 J) ischemia was induce IRI skeletal muscle. After 2 h (h) warm ischemic time (WIT) reperfusion, reporter protein expressing (TdTomato or Luci-GFP, 1 × 10 6 cells) obtained from isolated were injected intramuscularly. Surface marker expression potential analyzed vitro flow cytometry assay. In vivo bioluminescence imaging histopathologic evaluation biopsies performed quantify fate, Results 2h WIT induced severe on muscle, fiber regeneration as per histopathology within 14 days after injury. Bioluminescence demonstrated signals animals controls over the period (75 days). detected at injection site increased time. TdTomato myofibers visible host tissue postoperative 14, respectively, suggesting that differentiated fibers. Higher found compared without ischemia, indicative enhanced growth and/or IRI-affected antagonizing damage caused IRI. Conclusion WIT-induced requests numbers engraft persist, possible rational therapy antagonize Further investigations are needed evaluate capacity therapeutic advantage setting limb

Cytoglobin modulates myogenic progenitor cell viability and muscle regeneration.
Mammalian skeletal muscle can remodel, repair, and regenerate itself by mobilizing satellite cells, a resident population of myogenic progenitor cells. Muscle injury and subsequent activation of myogenic progenitor cells is associated with oxidative stress. Cytoglobin is a hemoprotein expressed in response to oxidative stress in a variety of tissues, including striated muscle. In this study, we...
متن کاملIntramuscular transplantation and survival of freshly isolated bone marrow cells following skeletal muscle ischemia-reperfusion injury.
BACKGROUND Delayed treatment cellular therapies offer an attractive means to treat extremity injuries involving acute skeletal muscle ischemia-reperfusion injury (I/R). Bone marrow is a rich source of stem and progenitor cells with the potential to improve skeletal muscle regeneration. The extent to which bone marrow cells (BMCs) may be useful for I/R is not known. The purposes of this study we...

OBJECTIVES To evaluate the effects of alprostadil (prostaglandin [PGE1] analog) and iloprost (prostacyclin [PGI2] analog) on renal, lung, and skeletal muscle tissues after ischemia reperfusion (I/R) injury in an experimental rat model. MATERIALS AND METHODS Wistar albino rats underwent 2 hours of ischemia via infrarenal aorta clamping with subsequent 2 hours of reperfusion. Alprostadil and il...
متن کاملUse of spleen organ cultures to monitor hemopoietic progenitor cell regeneration following irradiation and marrow transplantation.
After lethal irradiation of C57BL mice followed by the injection of 10(7) marrow cells, total cellularity and progenitor cell levels exceeded pretreatment levels within 12 days in the spleen, but regeneration remained incomplete in the marrow. The exceptional regenerative capacity of progenitor populations in the spleen was observed in organ cultures of spleen slices prepared 24 hr after irradi...
